Effect of histochrome, an antioxidant drug, on contractility and metabolism of isolated rat heart in "calcium paradox", ischemia and reperfusion

Abstract: A comparative study of protective effect of a new antioxidant histochrome on isolatedrat heart at calcium paradox and ischemia-reperfusion was carried out. Perfusion withCa-containing solution after 10-min perfusion with Ca-free medium caused depletion of highenergy phosphates, loss of myoglobin, uncoupling of mitochondria, increase of leftventricular diastolic pressure. Pretreatment with histochrome led to decrease ofcardiomyocyte damage evaluated by myoglobin efflux in perfused solution, attenuation ofdecrease ATP and phosphocreatine, and coupling of mitochondria. This resulted in adecrease of left ventricular diastolic pressure at calcium paradox. Less effect ofhistochrome treatment was noted in the case of ischemia-reperfusion model. It wasconcluded that histochrome effectively prevents heart from abnormalities caused by calciumimbalance in heart.
