Методы экспериментальной валидации потенциальных белков-мишеней для новых лекарственных средств

Год: 2005 том: 51 выпуск : 1 страницы: 2-18 Обзор посвящен описанию основных существующих и разрабатываемых технологий для экспериментальной валидации потенциальных белков-мишеней, предсказанных in silico методами сравнительной геномики и биоинформатики. Так как эта проблема на настоящий момент до конца не решена, то в обзоре приведено описание широкого круга методов, пригодных для валидации потенциальных мишеней.
В обзоре рассмотрены: (1) применение протеомных технологий (контроль экспрессии потенциальных белков-мишеней и их вариабельности, анализ белок-белковых взаимодействий); (2) геномные технологии в экспериментальной валидации мишеней (инактивация целевого гена, подавление транскрипции, разрушение целевой мРНК, подавление трансляции); (3) методы прямой инактивации целевого белка (моноклональные антитела, фотоинактивация белков, одноцепочечные и внутриклеточные антитела, аптамеры); (4) высокопроизводительные технологии; (5) валидация мишеней in vivo .Загрузить PDF:
