Antipeptide antibodies specific for the N-terminal site of lactate dehydrogenase from swine muscle

Antipeptide antisera were obtained after immunization of rabbits with N-terminal peptide of pig muscle lactate dehydrogenase-5 containing the enzyme antigen determinant. The N-terminal peptide (1-32 amino acids) was conjugated with bovine blood serum albumin. Specific antipeptide antibodies were isolated and purified using immunosorbent involving the antigen--lactate dehydrogenase-5. Immunodiffusion procedure and immunoenzymatic assay showed that the antipeptide antibodies obtained were specific towards conjugate, N-terminal peptide and the native pig muscle lactate dehydrogenase-5. The antipeptide antibodies towards N-terminal fragment of lactate dehydrogenase did not affect the enzymatic activity.
