Analysis of species differences of radioactively labeled ceruloplasmin and albumin by means of peptide maps

A procedure is developed for microanalysis of proteins, namely of human and rat ceruloplasmins as well as of their proteolytic fragments. The procedure involved electrophoretic purification of the peptides studied, labelling with 125I, tryptic hydrolysis and separation of the labelled peptides by cellulose thin-layer chromatography. Characteristics of the methods are described. Identification of individual polypeptide chains is reported. Specific localization of the labelled peptides on radioautogramms of the peptide maps was characterized. Analysis of human and rat ceruloplasmins, their fragments as well as human and rat blood serum albumins is presented as an example.
